Codevilla,Cristiane Franco; Tischer,Bruna; Gindri,Amanda Leitão; Nogueira-Librelotto,Daniele Rubert; Barin,Juliano Smanioto; Silva,Cristiane de Bona da; Rolim,Clarice Madalena Bueno; Zepka,Leila Queiroz; Menezes,Cristiano Ragagnin de.
ABSTRACT: The high intensity ultrasound-assisted extraction (HIU) is one of the most simple, quick and efficient techniques for the extraction of phenolic and other antioxidant compounds from plants. This is the first application of HIU for the extraction of these compounds from goldenberry fruit. The HIU and conventional extraction techniques showed similar results regarding to phenolic compounds and antioxidant capacity. However, the time required for HIU extraction (5min) was 24 times lower than conventional extraction (120min). Phenolic compounds reported were chlorogenic acid, caffeic acid and rutin. In vitro cytotoxicity assays were used for evaluation of extracts and the results showed that in a wide range of concentration, the extract maintains...

Cytotoxicity and genotoxicity of low doses of mercury chloride and methylmercury chloride on human lymphocytes in vitro BJMBR
Silva-Pereira,L.C.; Cardoso,P.C.S.; Leite,D.S.; Bahia,M.O.; Bastos,W.R.; Smith,M.A.C.; Burbano,R.R..
Mercury is a xenobiotic metal that is a highly deleterious environmental pollutant. The biotransformation of mercury chloride (HgCl2) into methylmercury chloride (CH3HgCl) in aquatic environments is well-known and humans are exposed by consumption of contaminated fish, shellfish and algae. The objective of the present study was to determine the changes induced in vitro by two mercury compounds (HgCl2 and CH3HgCl) in cultured human lymphocytes. Short-term human leukocyte cultures from 10 healthy donors (5 females and 5 males) were set-up by adding drops of whole blood in complete medium. Cultures were separately and simultaneously treated with low doses (0.1 to 1000 µg/l) of HgCl2 and CH3HgCl and incubated at 37ºC for 48 h. Genotoxicity was assessed by...

Cytotoxicity and genotoxicity of stilbene derivatives in CHO-K1 and HepG2 cell lines Genet. Mol. Biol.
Mizuno,Cassia Suemi; Ampomaah,Winnifred; Mendonça,Fernanda Ribeiro; Andrade,Gabriela Carvalho; Silva,Ariel Maria Nazaré da; Goulart,Mirian Oliveira; Santos,Raquel Alves dos.
Abstract The cytotoxicity and genotoxicity of the stilbenes (E)-methyl-4-(3-5-dimethoxystyryl)benzoate (ester), (E)-4-(3-5-dimethoxystyryl)aniline (amino), (Z)-1,3-dimethoxy-5-(4-methoxystyryl)benzene (cis-TMS) and (E)-1,3-dimethoxy-5-(4-methoxystyryl)benzene (trans-TMS) were investigated in this work. Structural modifications of resveratrol, a naturally occurring stilbene, have been previously performed, including the replacement of hydroxyl by different functional groups. Such modifications resulted in significant improvement of target-specific effects on cell death and antiproliferative responses. The parameters were evaluated using XTT assay, clonogenic survival assay and the cytokinesis-block micronucleus assay in CHO-K1 and HepG2 cell lines. The...

Cytotoxicity and genotoxicity of untreated hospital effluents BABT
Ortolan,Maria da Graça Silva; Ayub,Marco Antônio Záchia.
Untreated hospital effluent samples were tested for cytotoxic and genotoxicity in order to access whether they presented any serious health hazards. Results of umuC test suggested that the effluent from a large, general hospital could be considered weakly genotoxic. Comparatively, effluents from inpatient units presented higher frequency of genotoxicity than those coming from laboratory facilities. Although no cytotoxic activity was detected in laboratory effluent samples, cytotoxicity appeared to be an important problem of effluents from inpatient units.

Cytotoxicity and morphological analysis of cell death induced by Bothrops venoms from the northeast of Argentina J. Venom. Anim. Toxins incl. Trop. Dis.
Bustillo,S; Lucero,H; Leiva,LC; Acosta,O; Kier Joffé,EB; Gorodner,JO.
Bothrops snake venoms have been proved toxic to a variety of cell types, in both in vivo and in vitro models. Studies on the pharmacological actions of Bothrops venoms from Argentina are relatively scarce and the direct action of the crude venoms has not been assessed using cell culture models. In this work, we investigated the cytotoxicity of crude venoms from B. alternatus and B. diporus in a skeletal muscle (C2C12) cell line, which is commonly used as a model for studying the myotoxic action of snake venom. Both venoms (1.25-50 µg/mL) induced an early and significant decrease in cell viability. The cytotoxic concentration 50 (CC50), determined three hours after exposure, revealed that B. diporus venom was significantly more cytotoxic (CC50: 2 µg/mL)...

Geiger, Marie; Guitton, Yann; Vansteelandt, M.; Kerzaon, I.; Blanchet, Estelle; Du Pont, T. Robiou; Frisvad, J. C.; Hess, Philipp; Pouchus, Y. F.; Grovel, Olivier.
In order to assess the putative toxigenic risk associated with the presence of fungal strains in shellfish-farming areas, Penicillium strains were isolated from bivalve molluscs and from the surrounding environment, and the influence of the sample origin on the cytotoxicity of the extracts was evaluated. Extracts obtained from shellfish-derived Penicillia exhibited higher cytotoxicity than the others. Ten of these strains were grown on various media including a medium based on mussel extract (Mytilus edulis), mussel flesh-based medium (MES), to study the influence of the mussel flesh on the production of cytotoxic compounds. The MES host-derived medium was created substituting the yeast extract of YES medium by an aqueous extract of mussel tissues, with...

Cytotoxicity of chlorhexidine digluconate to murine macrophages and its effect on hydrogen peroxide and nitric oxide induction BJMBR
Bonacorsi,C.; Raddi,M.S.G.; Carlos,I.Z..
Chlorhexidine, even at low concentrations, is toxic for a variety of eukaryotic cells; however, its effects on host immune cells are not well known. We evaluated in vitro chlorhexidine-induced cytotoxicity and its effects on reactive oxygen/nitrogen intermediate induction by murine peritoneal macrophages. Thioglycollate-induced cells were obtained from Swiss mice by peritoneal lavage with 5 ml of 10 mM phosphate-buffered saline, washed twice and resuspended (10(6) cells/ml) in appropriate medium for each test. Cell preparations contained more than 95% macrophages. The cytotoxicity was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay and the presence of hydrogen peroxide (H2O2) and nitric oxide (NO) by the horseradish...

Rodrigues,M.; Barbosa Junior,F.; Perussi,J.R..
This paper describes the effect of dipyridamole (DIP) on the cytotoxicity of cisplatin in HEp-2 human larynx cancer cells in vitro and the nature of the interaction between cisplatin and dipyridamole. Cytotoxic assays were performed to obtain the IC50 for cisplatin. The cells were treated with 0, 20, 40, 80, 120 or 200 µM cisplatin, with or without a single concentration of DIP and incubated for 60 min at 37ºC and 5% CO2 for 3 days and then counted with a hemocytometer. The accumulation of cisplatin in the cells was measured by atomic absorption and fluorescence was used to determine the membrane binding constant of DIP. In the presence of 10, 20 and 30 µM DIP, the IC50 of cisplatin was reduced by 25, 60 and 82% in HEp-2 cells. Combination index analysis...
